As the host can I turn on refueling for the league for one race even though in an update refueling wasn’t available anymore.

You can turn it on but it will only apply from the start of the next season. I don't know about any update to remove refueling entirely.

In a word, No.

The three rules (2 compounds, refuelling & fastest lap) are locked in for an entire season and cannot be changed once the season has been generated, which happens immediately after the last race of a season.

So the rules for next season have to be selected by the host before the final race of the current season.
